There was such a precious small amount of the final hydrochloride salt that, by the time the needed build-up of dosage had been completed, there was just enough left for this single trial, which was conducted in South America. Based upon the volunteered comparisons to LSD and TMA, a potency for this compound has been published that states that it is 4x the potency of mescaline, or 4 M.U. The material must be re-synthesized, and re-evaluated with the now-accepted protocol. 

The phenethylamine homologue of TMA-6 is well known, but is virtually unexplored pharmacologically. The above benzaldehyde with nitromethane in glacial acetic acid containing ammonium acetate gave the appropriate beta-nitrostyrene as yellow crystals with a mp 177-177.5 °C. This, with LAH in ether, gave 2,4,6-trimethoxyphenethylamine (2,4,6-TMPEA, or 2C-TMA-6) as the picrate salt (mp 204-205 °C) or the hydrochloride salt (mp 234-235 °C). It has been shown not to be a substrate to the soluble amine oxidase from rabbit liver, a property it shares with mescaline, but whether it is or is not active in man is at present unknown. 

Conformational results are often sensitive to the input geometry used. For instance, Pullman at at. (68) carried out two PCILO calculations for mescaline, using the x-ray crystal geometries obtained for the hydrobromide and hydrochloride salts these salts exist in gauche and extended conformations, respectively.

Throughout the 1950s and 1960s, mescaline could be purchased from several chemical supply houses in the form of sulfate or hydrochloride crystals. In 1970, mescaline and peyote were "scheduled as part of a Comprehensive Drug Abuse Prevention and Control Act, which established penalties for possession, manufacture or distribution "a term of imprisonment of not more than 15 years, a fine of not more than 25,000, or both. Title 21 proscribes possession of "all parts of the plant presently classified as Lophophora williamsii, whether growing or not, the seeds thereof, any extract from any part of such a plant, every compound, salt, derivative, mixture or preparation of such a plant, its seeds or extracts. ... 

Early studies of mescaline, detailed in Kluver s book, generally involved doses of a fifth to a half of a gram. Shulgin puts the average dosage used in experimental investigations at between "300 and 500 mg of the sulfate salt, which is equivalent to 225-375 mg of the hydrochloride. ... 

Mescaline usually appears as a clear crystal in tbe form of a sulfate or hydrochloride. The crystals are up to lA" (1/3 cm.) long. Shulgin puts the usual dosage of the sulfate salt at 300-500 mg., which is equivalent to 225-375 mg. of the hydrochloride. He adds that this dose is often administered in two installments about an hour apart in order to minimize nausea.
