2-Mercapto-succinic acid

The same maleic acid macrolide was also used to produce bolaamphiphiles with two different head groups in high yield. For this purpose, the macrolide was first dissolved in 2-propanol and reacted with an alkaline solution of 2-mercapto-succinic acid. The solvent was then evaporated and the residue extracted with acetone to remove a minimal percent of the remaining macrolide. The product, which contained only one mercaptosuccinic acid substituent, was redissolved in hot 2-propanol/water 4 1 and then sodium bisulfite added. A bolaamphiphile with a large dicarboxylic acid head group and a small sulfonate head group was thus obtained in an almost quantitative yield . Of course, it is also possible to esterify the maleic acid with bolaamphiphile alcohols in order to obtain asymmetric, non-cyclic bolaamphiphiles (Scheme 2.10).

A positive aspect of this combinatorial scheme is typical for approaches with broad practical utility. Special building blocks (i.e., reactants belonging to a distinct chemical class with few commercially available analogues - in this case, mercapto-succinic acid) are used in a crucial role, namely for enabling the insertion of a diversity branch point allowing numerous subsequent derivatizations. 

Similar to biochip technology a poly-L-lysine monolayer can be bound via adsorption onto a gold cluster coated with a self-assembled monolayer of a carboxylated thiol e.g. mercapto-succinic acid. Using a biotinylated lysine-polymer proteins are immobilized via avidin conjugation. The poly-L-Iysine monolayer including all the proteins can be rinsed from the surface with a low or high pH buffer. 

Two 4-amino-3-mercaptol,2,4-triazole residues linked by a two-carbon bridge have been prepared by the reaction of thiocarbohydrazide with succinic acid 157a or malic acid 157b to give l,2-bis-(4-amino-5-mercapto-477-l,2,4-triazol-3-yl)ethan-l-ol 158a or l,2-bis-(4-amino-5-mercapto-477-l,2,4-triazol-3-yl)ethane 158b in yields of 56% and 60%, respectively (Equation 49) <2006PS(181)2361>.

Toxic heavy metals, such as cadmium, lead, and mercury, are sulfur seekers that bind strongly with thiol groups, which is one of the ways in which they interact adversely with biomolecules, including some enzymes. Advantage has been taken of this tendency to use thiols in chelation therapy in heavy metal poisoning.
