Maxi prep

Mouse caveolin-1, C-terminal fused to yellow fluorescent protein (YFP pEX EFl Cav-l-YFP ATCC). The plasmid is supplied in an Escherichia coli clone and can be propagated using standard bacterial propagation techniques with kanamycin as selection pressure. Purify the plasmid using a maxi-prep kit from Qiagen, Sigma or other vendors.

Plasmid DNA maxi prep kit, e.g., GeneHelute HP Plasmid Maxiprep Kit (Sigma Chemicals) or NucleoBond Xtra Maxi PLUS (Macherey-Nagel, Diiren, Germany). 

Isolate plasmid DNA from recombinant E. coli cultures following the procedure recommended by the manufacturer of the maxi prep kit. Concentrate the extracted DNA by alcohol precipitation. Alternatively use Nucleobond Finalizers (contained in the NucleoBond Xtra Maxi PLUS— follow the manufacturer s protocol). DNA should be concentrated at I pg/pL, ready for downstream applications. Prior to use, verify size and quality of plasmid DNA by agarose gel electrophoresis.
