With growing data banks of rate constants of elementary steps, it appears that kinetics assisted design of new catalytic processes will accelerate the development of improved catalytic processes. With powerful computational methods of thermodynamic and kinetic parameters, microkinetic analysis has emerged as a mature method to describe existing processes in combustion, polymerization, homogeneous and heterogeneous catalysis. It is reasonable that the process of vesiculation occurs at an equilibrium rate, constant for each cell species and modified by the metabolic demands of the cells. We have suggested (Verity and Brown, 1968b), from maturation studies of cerebral lysosomal enzymes, that for a given rate of vesiculation, the enzyme complement of the primary lysosome is directly proportional to the individual rates of synthesis of the component enzymes. Such a hypothesis would account for the heterogeneity in acid hydrolase reaction to injury, the difference in maturation profiles of individual acid hydrolases, and the dysynchrony of acid hydrolase induction. Also suggested is a direct feedback mechanism whereby a stimulation of vesiculation may in turn induce increased enzyme synthesis, possibly through changes in membrane phospholipid metabolism. [Pg.220]

The CCCBDB is currently the only computational chemistry or physics database of its kind. This is due to the maturity of quantum mechanics to reliably predict gas-phase thermochemistry for small (20 nonhydrogen atoms or less), primarily organic, molecules, plus the availability of standard-reference-quality experimental data. For gas-phase kinetics, however, only in the past two years have high-quality (<2% precision) rate-constant data become available for H and OH transfer reactions to begin quantifying uncertainty for the quantum mechanical calculation of reaction barriers and tunneling.
