Macrolide antibiotics absorption

Omura et al.21 used a reverse phase high performance liquTcT cEromatographic column, JASCO PACK SV-02-500, for macrolide antibiotics with methanol, M/15 acetate buffer pH 4.9, and acetonitrile (35 60 5) as solvent. A variable wavelength UV detector using the absorption of the individual compounds gave the required sensitivity. Alterations of buffer pH and the composition ratio of the mobile phase gave selectivity for separation of individual macrolide antibiotics. 

Macrolide antibiotics are administered orally, but many of them exhibit low and/or variable degrees of oral absorption, low serum concentrations, and short half-lives. Consequently, selection of semi-synthetic derivatives for development has often been guided by greater oral bioavailability, longer half-life, and higher and more prolonged concentrations in serum and tissues. Another antibiotic drug studied in presence of CD is natamycin (21, Fig. 7), a polyene macrolide antibiotic. Formation of inclusion complexes with fi-CD, HP-jS-CD or y-CD causes a very small red shift ( 1 nm) and band broadening of its UV absorption spectrum [75].
