Luminol enhanced chemiluminescence

The luciferin- and the luminol-enhanced chemiluminescence were measured by a multichannel luminescence analyzer (BIOLUMAT LB 9505, Berthold, Wilbad, Germany) in order to determine active oxygens liberated from leukocytes. Luciferin derivative and luminol are reagents unique to superoxide anion (Os") and hypochlorite ion (OCr), respectively. 

The oxygen burst of monocytes induced by ambient air particles (with the exception of oil fly ash particles) was less than the response elicited by polymorphonuclear leucocytes (Prahalad etal. 1999). The luminol-enhanced chemiluminescence response of polymorphonuclear leucocytes separated from heparinised human blood was generally increased with all washed particles, with oil fly ash and one urban air particle showing statistically significant (P <0.05) differences between dHjO-washed and unwashed particles. The luminol-enhanced chemiluminescence activity in polymorphonuclear leucocytes induced in post particles... 

In monocytes, luminol-enhanced chemiluminescence is an entirely extracellular event requiring both the release of myeloperoxidase and the generation of superoxide (Albrecht and Jungi 1992). 

Erythrocuprein inhibited the luminol-enhanced chemiluminescence caused by singlet oxygen produced in a xanthine-xanthine oxidase system as measured in a Packard scintillation counter (Weser and Paschen 1972). 

In isolated porcine leucocytes, 12-0-tetradecanoylphorbol-13-acetate produced a concentration-related luminol-enhanced chemiluminescence, that was markedly (P < 0.05) suppressed when the nitric oxide donor, pirsidomine was given intravenously 10 min before (Wainwright and Martorana 1993). 

Captopril inhibited (IC50 = 7.6 [xM) the opsonized zymosan-induced luminol-enhanced chemiluminescence of human blood polymorphonuclear neutrophils (Nagy et al. 1997). 

Aluminium salts stimulated luminol-enhanced chemiluminescence production by human neutrophils (Stankovic and Mitrovic 1991). Kong et al. (1992) described an Al(lll) complex with O which was a stronger oxidant than 02 " itself and which may contribute to the adverse biological effects of Al(III). Aluminium can enhance hydroxyl radical production by iron, but it is not in itself capable of catalysing the generation of hydroxyl radical fi-om the Fenton or Haber-Weiss reactions (Gutteridge et al. 1985). HL-60-M cells challenged by glass wool code A or stone wool code G fibres preincubated in unbuffered sahne for about 4 weeks, produced less intense luminol-enhanced chemiluminescence, as compared with freshly suspended fibres (Zoller and Zeller 2000). MMVF 21 and HT-N fibres as well as crocidohte and erionite did not show any decrease in chemiluminescence intensity after incubation in aqueous solutions.
