A site of a gene on a chromosome, i.e. site on a DNA molecule is called the locus and a variant at a particular locus namely a DNA sequence variant at the particular locus of interest, is termed an allele. What we observe, namely the complete set of observable inherited characteristics of an organism, is known as the phenotype, while what we obtain, namely the genetic constitution, which is the particular set of alleles inherited by the organism as a whole, is known as the genotype. Puff formation starts morphologically with the formation of a pale zone in the chromosome and disappearance of the precise outlines of the transverse strips and discs. The disc outlines then disappear and the chromosome starts to thicken in this area.
