Lignins distribution

An alternative approach to measurement of lignin distribution is to assess variation in the porosity of wood which has been hydrolyzed to form a lignin skeleton (Donaldson 1987). Hydrolysis with sulfuric acid is particularly useful because of the swelling that occurs. This magnifies areas of the cell wall such as the S3 layer or the compound middle lamella which would otherwise be too small to measure. [Pg.127]

Three quantitative approaches to measure lignin distribution are interference microscopy, UV microscopy, and bromination/EDXA. The choice of method probably depends more on the availability of equipment than any other factor since each method can be applied to a similar range of specimen types. [Pg.128]

All values for interference microscopy have been converted from v/v to w/w using density values for lignin and carbohydrate of 1.335 and 1.521. (Stamm and Sanders 1966)... Table 4.3.1. Interference microscopy can be applied to the study of lignin distribution in wood, in differentiating xylem, and in chips during pulping. The only constraints are that the material can be embedded and sectioned, matched samples are available for calibration, and the specimen can be oriented to obtain transverse sections. [Pg.129]
