L-Ethoxy-2-propanol

Base-catalyzed epoxide opening is a typical S -2 reaction in which attack of the nucleophile takes place at the less hindered epoxide carbon. For example, 1,2-epoxypropane reacts with ethoxide ion exclusively at the less highly substituted, primary, carbon to give l-ethoxy-2-propanol.

Trifluoroacetaldehyde, generated from l-ethoxy-2,2,2-trifluoroethanol (4 mmol) by treatment with polyphosphoric acid (PPA), was passed into a toluene solution of (Ry BIN0L-Ti(0-/-Pr)2 (1 mmol) at -78 °C in the presence of 2-propanol (2 mmol) coproduced during the preparation of the catalyst from Ti(0-/-Pr)4 and (i )-binaphthol (BINOL). The resulting hemiacetal was treated with benzoyl chloride, triethylamine and 4-dimethylaminopyridine (DMAP) at -78 °C to afford 2,2,2-trifluoro-l-isopropoxyethyl benzoate (80% ee) in 85% yield. When the esterification was carried out at higher temperatures, % ee of the product considerably decreased due probably to racemization of the intermediate hemiacetal. 

There are five additional five-carbon multifunctional ethers that have been the subject of a single study of their OH reactivity. These include l-ethoxy-2-methoxyethane (Dagaut et al., 1989a), 3-methoxy-1-butanol (Dagaut et al., 1989b), 3-ethoxy-l-propanol (Dagaut et al., 1989b), 2-isopropoxyethanol (Porter et al., 1997), and 1,3-dimethoxypropane (Moriarty et al 2003). For the 1,3-dimethoxypropane relative rate study of Moriarty et al. (2003), the rate coefficient data of Porter et al. (1997) have been used for the reference reaction (OH - - 2-ethoxy ethyl ether, see section ni-C-14 for details). Data are summarized in table III-C-15. In the absence of corroborative measurements, uncertainties of 35% are estimated for these rate coefficients.
