Electrophoretic and kinetic studies of the patient s enzyme have been reported in several cases (F10). Most of them showed decreased substrate affinity and abnormal electrophoretic mobility. The main cause of P5N deficiency is considered to be an abnormality of P5N-I, probably arising from a structural gene mutation (H6). The precise molecular defect has not been clarified, because the normal gene for P5N-I has not been isolated.

In 1961 Ansorge et al. (A5), using the same technique as previously, determined the peptide composition of urine derived from four normal subjects, three males and one female. Among twenty isolated peptides, seventeen were found in all specimens of urine, two peptides in three specimens, and the remaining one only in two specimens of urine. The identity of individual peptides was established on the basis of their electrophoretic and chromatographic behavior, as well as the amino acid composition after complete hydrolysis. It should be pointed out, however, that the amino acid composition of the peptides examined differs considerably from that obtained by the same authors in the case of the peptides described in 1958 (HI).

An alternative to nucleic acid isolation by electrophoresis followed by electrophoretic elution is HPLC. There are several advantages in the use of HPLC. It is simple and easy to use and has a high degree of resolution, it is faster than electrophoresis and contamination of DNA fragments by impurities from the agarose gel is avoided.
