Immunoglobulin-gold conjugates

Immunoglobulin-gold conjugates host anti-species-specific IgG coupled to 10 or 15 nm gold particles (e.g.. Sigma Chemicals). 

Incubate grids for 30 min at room temperature in colloidal gold conjugated with protein A, protein G, immunoglobulin, or if a biotinylated secondary antibody was used, streptavidin (see Note 6). 

Sodium azide inhibits peroxidase enzyme, therefore it should be left out from the diluents used with PO-immunoglobulin conjugates. Add 0.05% Tween-20 when diluting gold reagents. 

The gold nanoparticle/polyelectrolyte coated latex particles are then conjugated with biotin molecules through a layer of biotinylated poly(allylamine hydrochloride) that is deposited on the particle surface before biotin binding.
