Immune labeling

Key Words Transmission electron microscopy scanning electron microscopy Gram-positive Gram-negative immune labeling histochemistry colloidal gold quantum dots correlative microscopy fluorescence. [Pg.173]

Immunolabelling of thylakoid polypeptides was carried out following SDS-PAGE (see 2.4) and electroblotting of proteins to nitocellulose paper [9]. The protein blots were incubated with anti- antibody at 1 400 dilution and immune labelled bands visualised with peroxidase-second antibody conjugate plus diaminobenzidine/H202 as substrate. [Pg.1742]

A cascade of proteins of the immune response that can be triggered by antigen-antibody complexes and by the innate immune system (e.g. exposure to microbial polysaccharides) to raise the immune response. Complement proteins can detect and bind to foreign material or immune complexes and label them for phagocytosis. They can also cause inflammation by directly degranulating mast cells and releasing chemokines to recruit other immune cells into the affected area. [Pg.385]

In-vitro models can provide preliminary insights into some pharmacodynamic aspects. For example, cultured Caco 2 cell lines (derived from a human colorectal carcinoma) may be used to simulate intestinal absorption behaviour, while cultured hepatic cell lines are available for metabolic studies. However, a comprehensive understanding of the pharmacokinetic effects vfill require the use of in-vivo animal studies, where the drug levels in various tissues can be measured after different dosages and time intervals. Radioactively labelled drugs (carbon-14) may be used to facilitate detection. Animal model studies of human biopharmaceutical products may be compromised by immune responses that would not be expected when actually treating human subjects. [Pg.64]

Figure 9. Anti-PbTx antiserum inhibition of [ H]PbTx-3 binding to its receptor site in rat brain membrane preparations. Labeled toxin (0.5 nM in 1 ml PBS) was incubated with rat brain membranes (125 fig total protein) and increasing amounts of anti-PbTx antiserum (- -) or pre-immune serum (- -) for 1 hr at 4 C. Membrane-bound radioactivity was then measured in a centrifugation assay as previously described (8),... Figure 9. Synthetic hCT Is not commercially available now. Most assays have been developed by using hormone supplied as gifts from the CIBA-Gelgy Co., Basel, Switzerland for 131i or labeling. Immunization and standards although several useful antisera have been produced by Immunization with crude extracts of medullary carcinoma tissue. [Pg.51]

Figure 18.14 NHS-SS-PEG4-biotin can be used to label a primary antibody molecule that has specificity for a protein or interest. Incubation of the biotinylated antibody with a sample, such as a cell lysate, allows the antibody to bind to its target. Capture of the antibody-antigen complex on an immobilized streptavidin reagent effectively isolates the targeted protein from the other proteins in the sample. The disulfide linkage in the spacer arm of the biotin tag permits elution of the immune complex from the streptavidin support using DTT and without using the strong denaturing condition typically required to break the streptavidin-biotin interaction.
