Image reconstruction techniques

Two-dimensional periodic organization of eukaryotic and prokaryotic ribosomes occurs under special conditions in vivo (Byers, 1967 Kress et al., 1971 Taddei, 1972 O Brien et al, 1980) or in vitro (Barbieri, 1979 Clark et al, 1982). The two-dimensional sheets have been analyzed by image-reconstruction techniques (Kiihlbrandt and Unwin, 1982 Clark et al, 1982).

Likewise, advances in electron cryomicroscopy and image reconstruction techniques allow time-resolved investigations of structural transitions associated with capsid assembly and maturation (Conway et al., 2001 Lawton et al, 1997). These developments have been paralleled by refinements in the molecular approaches used for sample preparation, with the result that synthesis of assembly intermediates and end products has become routine for many viruses.

Compared to the structure of the reaction center of purple bacteria, the knowledge about Photosystem II is still in a very early stage. As a first step in the unravelling of the structure of PS II, we report in this communication the preparation of two-dimensional crystals of the PS II reaction center material comprised of CP47-Dl-D2-cytochrome b559, and of the subsequent analysis of these crystals by electron microscopy and image reconstruction techniques.

The transverse images of cold and hot regions insert were obtained by the filtered back projection image reconstruction technique. It was corrected for uniformity and center of rotation (COR). Chang s attenuation correction method was applied by choosing 0.111, 0.121, 0.131, 0.141 and 0.151 cm linear attenuation coefficient values. Butterworth filter with cut off frequency 0.35 cycles/cm and order 5 was used. Scatter correction technique was not applied due to non-availability with this gamma camera system.
