The efficacy of aliskiren monotherapy and aliskiren in combination with the diuretic hydrochlorothiazide (HCT), aliskiren in combination with the angiotensin receptor blocker (ARB) valsartan, and aliskiren in combination with the ACE inhibitor ramipril in reducing mean sitting diastolic blood pressure (msDBP) have been reported (Table 3). Manna et al. described a rapid and accurate reversed-phase ion-paring liquid chromatographic method, with ultraviolet detection, for the simultaneous assay of benazepril hydrochloride, fosinopril sodium, ramipril and hydrochlorothiazide [32], The separation was achieved on a LC-8 (12.5 cm x 4 mm, 5 pm particle size) column. The mobile phase consisted of 20 mM sodium heptansulfonate (pH 2.5) and methanol (8 17 v/v). Validation of the method showed it to be precise, accurate, and linear over the concentration range of analysis, with a limit of detection of 2 ng for benazepril. The method was applied to the analysis of three different binary commercial formulations.

In the first edition of this book, I wrote of response surface designs as applied in clinical research that the field is still in its infancy . In the second edition I am tempted to report, flippantly, that the infant in question does not seem to have grown much. However, an interesting paper that reports an actual clinical trial is that by Tangen and Koch (2001). They consider an example of a 3 x 4 factorial trial in hypertension in which on average 43 patients were randomized to each of 12 possible combinations of three levels of hydrochlorothiazide (placebo, low, high) and four levels of an ACE inhibitor. (For a remarkably similar trial reported in the medical literature, see the 3x4 factorial trial of felodipine and ramipril in 507 patients reported by Scholze et al. (1999).)...
