Coenzyme Q4 (Ubiquinone-4, 2,3-dimethoxy-5-methyl-6-[3,7,ll,15-tetrametbyl-hexadeca-2/,6/,10/,14-tetraenyl]-[l,4]benzoquinone [4370-62-l]M 454.7, m 30 , 33-45 , A (275nm) 185. A red oil purified by TLC chromatography on Si02 and eluted with Et20-hexane. Purity can be checked by HPLC (silica column using 7% Et20-hexane). It has A- ax 270 nm (e 14,800) in pet ether. [NMR and MS Naruta J Org Chem 45 4097 1980 cf Morton Biochemical Spectroscopy (Adam Hilger, London, 1975) p 491]. It has also been dissolved in MeOH/EtOH (1 1 v/v) and kept at 5 until crystals appear [Lester and Crane Biochim Biophys Acta 32 497 1958]. [Pg.522]

The TLC process is an off-line process. A number of samples are chromatographed simultaneously, side-by-side. HPTLC is fast (5 min), allows simultaneous separation and can be carried out with the same carrier materials as HPLC. Silica gel and chemically bonded silica gel sorbents are used predominantly in HPTLC other stationary phases are cellulose-based [393]. Separation mechanisms are either NPC (normal-phase chromatography), RPC (reversed-phase chromatography) or IEC (ion-exchange chromatography). RPC on hydrophobic layers is not as widely used in TLC as it is in column chromatography. The resolution capabilities of TLC using silica gel absorbent as compared to C S reversed-phase absorbent have been compared for 18 commercially available plasticisers, and 52 amine and 36 phenolic AOs [394]. [Pg.221]

The main limitation of these CSPs is their limited pressure stability, which makes them not very suitable for HPLC application. However, they have proved to be an excellent tool for the preparative separation of drugs by low-pressure HPLC. To make these CSPs accessible to HPLC, silica gel-based phases were developed. " This type of phase is available from Merck (Darmstadt, Germany) under the name Chiraspher. Polymer phases of different types have been developed by Okamoto s group. > They are prepared by the asymmetric polymerization of triphenylmethyl-methacrylate monomers. The original character of these polymers is that they do not possess any chiral centre and therefore their chirality is only due to their helicity. However, clear mechanisms have not been proposed... [Pg.476]

A solution of N-(4-pyridinyl)-lH-indol-l-amine (6 g) in 25 ml of dimethylformamide was slowly added to an ice-cooled suspension of NaH (1.3 g of 60% NaH dispersion in mineral oil was washed with hexanes, the liquid was decanted and the residual solid was dispersed in 5 ml of dimethylformamide). After anion formation, a solution of 1-bromopropane (4 g) in 5 ml of dimethylformamide was added. After one hour of stirring at ambient temperature, the reaction mixture was stirred with ice-water and extracted with dichloromethane. The organic extract was washed with water and saturated sodium chloride solution, was dried over anhydrous magnesium sulfate, filtered and concentrated to 8 g of oil. This oil was purified by HPLC (silica, ethyl acetate) and thereafter by column chromatography (alumina, ether) to give 6.4 g oil. This oil was converted to the maleate salt and recrystallized from methanol/ether to give 6.8 g of crystals, m.p. 115-116°C. [Pg.600]
