Horseradish peroxidase mimics

Nolte et al 46) produced an artificial enzyme based on the T4 replisome and applied it to the epoxidation of double bonds in synthetic polymers. Smith et al 51) reported that horseradish peroxidase catalyzes the oxidative polymerization of glucuronic acid. In recent literature, many biomimetic macromolecules with enzyme-like structures or functions have been reported including those that are dendrimers 64-66), those that have specified three-dimensional structures or recognition elements created by molecular imprinting 67), and other enzyme mimics 68). 

As briefly summarized in Section 1.2.2.1, extensive evidence has been reported indicating that horseradish peroxidase Compound I is an oxo-ferryl [Fe =0] porphyrin n-cation radical and that Compound II is an oxo-ferryl porphyrin. Groves and co-workers have reported an inorganic model complex for Compound I [53, 54] and Balch and co-workers have described a Compound II model [55, 56]. These models each appear to have the expected compositions for the respective enzyme states that they are designed to mimic. 

The other major class of DNAzyme reported mimics the action of enzymes like horseradish peroxidase, utilising in the process hydrogen peroxide as oxidant. Various reports describe mechanistic details of this class of DNAzyme, and a number of modifications have also been described. These include the modification by addition of PEG to the DNAzyme to make the DNAzyme active in organic solvents, the effect of backbone modifications, and a variant of the DNAzyme that requires Pb(II) ions for activity. A variant of the DNAzyme has been reported that mimics a NADH oxidase, and variants are reported that act as logic-controlled biofuel cells, " " and for the generation of DNA nanostructures. Publications have been described that use the peroxidase DNAzyme as a colourimetric assay for the detection of PCR products, for a fluorimetric assay of genetically-modified...
