Hemoglobin analysis

Reports describing patients with Hb-S-/3-thalassemia or with Hb-C-)3-thalasseraia are numerous [ (A5, A6, B28, C22, K21, K23, N12, R38, S50, S51, S63) and many others]. The patients suffer from a mild to rather severe anemia (Hb levels between 7.5 and 12 g/100 ml) which is usually microcytic and hypochromic. Hemoglobin analysis shows a high percentage of the abnormal variant (60-80%), elevated Hb-A2 levels (4-7%), variable amounts of Hb-F (1-20%) and of Hb-A (0-20%). Patients who produce notable amounts of Hb-A are usually less severely affected than patients in which the normal Hb-A production is completely suppressed. [Pg.190]

Basic information on a-thalassemias, i.e., defects with an impaired production of a polypeptide chains, is still limited because of difficulties in identifying the simple adult heterozygote. There are a range of applications of conventional cIEF to real-world analysis, including the characterization of protein samples in laboratories of biotech pharmaceutical companies, and clinical analysis. In the pharmaceutical industry, some methods have been developed based on this technology [13,14] and validated [15-17] as the identity assays in regulated pharmaceutical laboratories for the analysis of therapeutic monoclonal antibodies and glycoproteins. cIEF has also been used in clinical analysis for human hemoglobin analysis [18,19] and the analysis of proteins in cerebrospinal fluid [20]. There are several reviews on the applications of the conventional cIEE [10,21]. However, the pace of acceptance of conventional cIEF for the analysis of real-world samples is slow. [Pg.566]

Although CE instruments are not well designed for CIEF, many variants can be separated better by this technique. In addition to the common variants, G Philadelphia, A2, and Bart s can all be separated by CIEF [46-48]. The separation by CIEF compares well with gel isoelectric focusing and with HPLC. The variants have also been analyzed by both CE and CIEF equipped with special absorption imaging detectors. These types of detection devices eliminate the extra steps needed to move the peaks, after the focusing step, to the detector and can simultaneously detect several capillaries with better precision and faster results than CE instruments.
