Haptens oligosaccharide

Immobilization of A and B blood group oligosaccharide haptens and preparation of immunoadsorbents with specificity to anti-A and anti-B antibodies has been carried out with the use of poly acrylate-coated PG (WPG-PA) [124]. Prespacered A and B-trisaccharide-fl-aminopropylglycosides were used for the synthesis. WPG-PA (1 g) quantitatively binds both haptens (2 pinole) whereas some other activated affinity supports (for example, CNBr-Sepharose 4B) do not. On the other hand, glycidoxypropyl-silica binds prespacered haptens completely but these materials reveal no specific adsorptivity.

The specificity of the antiserum raised against the Lea neoglycoprotein (9) was determined by hapten-inhibition studies using synthetic oligosaccharides,72 and derivatives of bovine and horse serum-albumin to which the Lea trisaccharide had been attached.73 On the basis of hapten inhibition of precipitation, this antiserum was shown to be highly specific for the carbohydrate portion of the synthetic antigen,... 

Add the aldehyde-containing oligosaccharide to the carrier solution at a concentration sufficient to obtain at least a 20-fold molar excess of hapten to carrier. 

Within the Cl oligosaccharide hapten it is likely that several different regions elicit complementary antibodies. Hopefully, further structural studies and the synthesis of a few oligosaccharides may help us identify an 07 antigenic determinant with even higher specificity for Salmonella serogroup Cl. 

Clearly, the antibodies formed against the oligosaccharide hapten in this case recognize the modified glucose linkage arm present in the sugar derivative used to prepare the immunogen. Why the antibodies produced are specific for the reduced...
