Many variants of the two-hybrid system have been devised.d For example, a green fluorescent protein reporter can be used/ Because significant biological protein-protein interactions often require that three or more proteins interact,d hybrid systems involving more than two proteins have been developed. Two-hybrid systems for bacteria have also been devised.1" A virtue of the two-hybrid methods is that they work with undenatured, if not totally natural, proteins. This is in contrast to widely used methods that involve separation of denatured proteins on gels or columns. 

The entire three-dimensional biofilm structure was imaged using MRI. This was then correlated to a fluorescent CLSM image by employing a green fluorescent protein reporter construct of S. oneidensis. Non-invasive techniques which enable measurements of dynamic metabolic processes, especially in a depth-resolved fashion, are expected to advance our understanding of processes occurring within biofllm communities. 

Quantum yield of luciferin. Various values of quantum yield have been reported for coelenterazine in the luminescence reaction catalyzed by Renilla luciferase 0.055 (Matthews et al., 1977a), 0.07 (Hart, et al., 1979), and 0.10-0.11 (with a recombinant form Inouye and Shimomura, 1997). The quantum yield is significantly increased in the presence of Renilla green fluorescent protein (GFP) see below.

GFP has also been proposed as a successor to the Ames and SOS chromotest. Billinton et al. [8] obtained a reporter system, employed as genotoxicity biosensor, that uses eukaryotic cells (the baker yeast Saccharomyces cerevisiae) instead of bacteria. The strain produces green fluorescent protein, codon optimized for yeast, when DNA damage has occurred. It was demonstrated that the reporter does not falsely respond to chemicals that delay mitosis, and responds appropriately to the genetic regulation of DNA repair.

It is recommended to linearize the plasmid before transfection to avoid aberrant integration of Tet-repressor coding sequence. Alternatively the construct can be delivered by viral-mediated infection. In this case use pLenti6/TRto express the repressor. It is recommended to optimize the transfection conditions by using an easily detectable reporter system such as green fluorescent protein (GFP).
