Among several thiol compounds, glutathione may provide neuroprotection in patients treated with cisplatin without altering its antineoplastic activity. This protective role may be based on blockade of the accumulation of p53 protein in response to platinum in dorsal root ganglia, thereby hindering platinum-based apoptosis (57,112).

The proposed activation of acetylhydrazine involves A-hydroxylation, followed by loss of water to yield acetyldiazine, an intermediate which would fragment to yield acetyl radical or acetyl carbonium ion (figure 7,15). Glutathione was not depleted by hepatotoxic doses of acetylhydrazine, indicating that unlike bromobenzene or paracetamol toxicity, it does not have a direct protective role.
