All glycolytic intermediates derived from glucose are phosphorylated. At physiological pH values, phosphate groups are ionized so that each intermediate is negatively charged. Charged molecules are not readily able to cross cell membranes unless they have a specific transport protein, so the intermediates are confined to the cytoplasm of the cell.
