Glibenclamide clinical studies

Numerous pharmaceutical companies have created Pgp modulators by modifying existing drugs such as verapamil, cyclosporine A, glibenclamide, and others from certain related chemical structures. These research efforts are in various phase I, II, and III clinical studies [214]. No single chemical structure has proven to be effective in phase III studies to date.

In a retrospective study 400 doctors in Germany were asked to compare the characteristics of severe hypoglycemia in patients in acute care induced by glimepiride and glibenclamide (49). Only 24 responded (6%). There were no differences in clinical characteristics or time course. 

The manufacturer notes that the concurrent use of moxifloxacin and glibenclamide resulted in an approximate 21% decrease in the peak plasma level of glibenclamide in diabetic subjects, but this did not alter blood glucose and endogenous insulin. A pooled analysis from clinical and postmarketing studies suggested that moxifloxacin had no clinically relevant effect on blood glucose homoeostasis, even in patients with diabetes mellitus."... 

Neither cimetidine nor rifampicin had any clinically relevant effect on the pharmacokinetics of nicorandil. Nicorandil did not alter the anticoagulant effects of acenocoumarol. Although animal studies surest antagonism of effects, a study in patients found no pharmacodynamic interaction between nicorandil and glibenclamide. Nicorandil may potentiate the hypotensive effects of other vasodilators, tricyclic antidepressants and alcohol.
