The proper preparation of a mull or paste is an excellent way to get a sample ready for qualitative infrared spectroscopy. One grinds vigorously about 3 to 10 mg of substance with a hard pestle in a hard and smooth mortar (e.g., agate) for 1 to 5 minutes until the powder is so fine that its caked surface takes on a glossy appearance. Just as a voyage from one point to another consists of many individual steps, so also does an analytical procedure involve many individual operations. These are often referred to as "unit operations", and they may be classified in several ways (eg. obtaining a sample, getting it ready for analysis, the analysis itself, or the detection step for quantitation). The sample may be solid, liquid, or gas, and its pretreatment could include such operations as extraction, filtration, dialysis, distillation, concentration, or even chromatography. 

When all the samples from one individual are ready, they are best packed in a cardboard box for the protection of the containers, and then placed in a polythene bag to ensure that the box does not get wet during transit to the laboratory. Relevant hazard warning labels should always be placed on the bag. At no time during the sampling procedure should there be any eating, drinking, or smoking. 

The quality of the DNA introduced into PCR is of critical importance. Highly purified template DNA and the absence of inhibitors are the best guarantee of enormous quantities of amplified products. Thus putting a lot of effort into this step of the analysis is highly recommended. Today, a variety of extraction protocols are available from different suppliers, including all components required to get sufficient amounts of DNA from various samples. In particular, for bacterial suspensions and for mammalian cell culmres, ready-to-use kits are on the market. During the investigation, it was discovered that a maintenance worker had found water in tank 701. He was told to check the Unit 7 tanks to ensure they were ready for the T34 production startup. Unit 7 had been shut down previously (see Physical Plant Context ). The startup was scheduled for 10 days after the decision to produce additional K34 was made. The worker found a small amount of water in tank 701, reported it to the maintenance manager, and was told to make sure the tank was bone dry. However, water was found in the sample taken from tank 701 right before the uncontrolled reaction. It is unknown (and probably unknowable) whether the worker did not get all the water out or more water entered later through the same path it entered previously or via a different path. We do know he was fatigued and working a fourteen-hour day, and he may not have had time to do the job properly. He also believed that the tank s residual water was from condensation, not rain. No independent check was made to determine whether all the water was removed.
