Gamma globulin data

A definitive identification of the proteins in each peak is not possible, however, the elution times of the peaks at 13-14 min. and 15 min. are close to the times which would be expected for gamma-globulins and albumins, two of the principal classes of serum proteins. These data also indicate the loading capacity of this column with serum. More than 14 mg. of undiluted serum was injected before evidence of overloading in the form of band broadening and peak distortion was observed.

Prepare a standard calibration curve using the data obtained for standard gamma globulin (Table E4.2, tubes 2 to 6). Plot the absorbance on the j axis and mg of standard protein per assay on the x axis. Use the standard curve to calculate the protein concentration in the isolated a-lactalbumin fractions (in units of mg/mL.) Compare your graph with Figure 2.6. 

In conjunction with the preceding observations and in view of the additional data reported below, it seems reasonable to term the material being studied a thyroid-stimulating gamma-globulin.
