After using this control step, no labeling should be seen but normal mouse anti-Ag B should be seen. [Pg.135]

The second control is the No second 1° labeling (Table 12.4) with mouse anti-Ag B 1 antibody and should show that the normal mouse IgG blocks all the labeled Fab fragment binding. Perform this control using the pattern established above. To the anti-mouse Fab 555 fluorophore, add the nonspecific IgG normal mouse serum and then add the mouse anti-Ag B 1° antibody. Next, mix the anti-Ag A and the anti-Ag B 1° antibodies and incubate with the cells. Again, no labeling should be seen when this solution is incubated with samples. This control must be done to confirm that the amount of blocking is still sufficient, especially if the ratio of labeled Fab to P antibody has been altered. [Pg.135]
