S. cerevisiae is produced by fed-batch processes in which molasses supplemented with sources of nitrogen and phosphoms, such as ammonia, ammonium sulfate, ammonium phosphate, and phosphoric acid, are fed incrementally to meet nutritional requirements of the yeast during growth. Large (150 to 300 m ) total volume aerated fermentors provided with internal coils for cooling water are employed in these processes (5). Substrates and nutrients ate sterilized in a heat exchanger and then fed to a cleaned—sanitized fermentor to minimize contamination problems. 

Plate and frame exchangers are suited to high sanitation services, and are 25-50% cheaper in stainless construction than shell-and-tube units.

For the implementation of techniques such as step-elution, gradient-elution, displacement-elution, as well as for the wash and sanitation steps needed for ion exchange chromatography and continuous downstream processing in general. 

The tanks and the loops must be sanitized NLT 80°C. The DIW is heated at 80°C and maintained at this temperature for 1 h once a day, during the night. After 1 h at 80°C, the loop must be cooled at 20°C by the cold exchanger located at the return of the loops. 

Storage tank and in the circulation loop at ambient temperature most of the time (this requires an extra heat exchanger with cooling water on the shell side). To achieve microbial control, such system (or any system that includes ambient loop) needs to be periodically sanitized by heating up its contents to about 80° C and holding the temperature for at least 1 hr. The sanitization frequency can vary from once a day to once a week or less the lower the frequency is, the more attention to the studies proving the effectiveness of the microbial control shall be expected. 

In the food and dairy industries, the sanitation and cleaning of the plant are extremely important. Here, the easy access to each membrane is very valuable for the inspection as well as an immediate exchange of broken membranes, securing optimum bacteriological conditions.

Preuse validation of deionizing systems used to produce purified water should include consideration of such factors as microbial quality of feed water (and residual chlorine levels of feed water where applicable), surface area of ion-exchange resin beds, temperature range of water during processing, operational range of flow rates, recirculation systems to minimize intermittent use and low flow, frequency of use, quality of regenerant chemicals, and frequency and method of sanitization.
