During tapping AFM, the cantilever oscillation amplitude remains constant when not in contact with the surface. The tip is then carefully moved downward until it gently taps the surface. As the tip passes over an elevated surface feature, the cantilever has less room to oscillate, and the amplitude of oscillation decreases (vice versa for a surface depression). The oscillation frequency is typically 50-500 kHz, with an amplitude of ca. 30 nm, which is sufficient to overcome the adhesive forces that are evident in contact (and noncontact) modes (Figure 7.49). Thermodynamic parameters for the mixing of dimyristoyl lecithin (DML) and dioleoyl lecithin (DOL) with cholesterol (CHOL) in monolayers at the air-water interface were obtained by using equilibrium surface vapor pressures irv, a method first proposed by Adam and Jessop. Typically, irv was measured where the condensed film is in equilibrium with surface vapor (V < 0.1 0.001 dyne/cm) at 24.5°C this exceeded the transition temperature of gel liquid crystal for both DOL and DML. Surface solutions of DOL-CHOL and DML-CHOL are completely miscible over the entire range of mole fractions at these low surface pressures, but positive deviations from ideal solution behavior were observed. Activity coefficients of the components in the condensed surface solutions were greater than 1. The results indicate that at some elevated surface pressure, phase separation may occur. In studies of equilibrium spreading pressures with saturated aqueous solutions of DML, DOL, and CHOL only the phospholipid is present in the surface film. Thus at intermediate surface pressures, under equilibrium conditions (40 > tt > 0.1 dyne/cm), surface phase separation must occur. [Pg.174]

Cholesterol and lecithin form completely miscible solutions in mono-layers at very low surface pressures, characterized by excess positive heats and excess negative areas of mixing. A dark control should always be placed alongside the authentic samples to compensate for temperature and humidity effects. However, such a reference sample will only describe thermal reactions occurring from the ground state of the molecule and not take into account thermal reactions from its excited state, i.e., initiated by a photochemical process. A reference sample wrapped in aluminum foil would not necessarily experience the same surface heating effect as the exposed sample due to reflection (i.e., blocking of the surface heating).
