Electrophoretic heterogeneity

Electrophoretic heterogeneity of whole antibody or even of active Fab proteolytic fragments is still an issue in the development of noncompetitive assay. To get a uniform peak in CE, Hafner et al. [29] proposed using singlechain antibody variable-region fragments (scFv), which can be produced by recombinant technology.

Many enzymes are glycoproteins, and variations in carbohydrate side chains are a common cause of nonhomogeneity of preparations of these enzymes. Some carbohydrate moieties, notably h/-acetylneuraminic acid (sialic acid), are strongly ionized and consequently have a profound effect on some properties of enzyme molecules. For example, removal of terminal sialic acid groups from human liver and/or bone alkaline phosphatase with neuraminidase greatly reduces the electrophoretic heterogeneity of the enzyme.

We have purified HGPRT to homogeneity from non-pooled, human male erythrocytes and have attempted to elucidate the nature of the electrophoretic heterogeneity of HGPRT activity observed during the purification (Arnold and Kelley, 1971 Kelley and Arnold, 1973).
