Drug binding: protein polymorphism

ABCBl is a 170-kDa integral membrane glycoprotein. It consists of 12 transmembrane helices and 2 nucleotide binding domains. There is no disease known that is linked to a nonfunctional protein due to mutations within the ABCBl gene, so far. In recent years, many screening studies with human individuals have been performed on the association of ABCBl polymorphisms with ABCBl expression and function in tissues and with the pharmacokinetics and pharmacodynamics of drugs [64]. However, still there are discrepancies in the results, and furthermore, no... [Pg.305]

Looking forward, advances in molecular biology and genetic engineering will lead to further discoveries of how drugs and metabolites are transported by the kidney and Uver. This will undoubtedly involve characterization of the substrate binding site of transport proteins and the mechanisms involved in transporter induction and inhibition. Furthermore, the role of genetic polymorphisms on individual transport systems will become better defined. [Pg.197]
