Almost as many methods exist for preparing cuticles for light microscopy as there are Drosophila biologists. In this chapter, we offer some simple protocols that we use routinely and have found satisfactory for analysis of fine cuticular details. With experience, investigators will undoubtedly evolve their own variations.
