Dopamine dopaminergic agents

In patients with hypothyroidism caused by hypothalamic or pituitary failure, alleviation of the chnical syndrome and restoration of serum T4 to the normal range are the only criteria available for estimating the appropriate replacement dose of levothyroxine. Because chronic cocaine use appears to reduce the efficiency of central dopamine neurotransmission, a number of dopaminergic compounds, including amantadine, bromocriptine, mazindol, and methylphenidate, have been examined as treatments for cocaine abuse. The close structural similarity of the semirigid conformational analogue 1 to the trans-rotamer of dopamine (DA) was noted almost ten years ago (1,2). The synthesis of 1 has been described more recently and its pharmacological profile determined (3-6) to be similar to alpha-methyldopamine (AMDA) which is inactive as a dopaminergic agent.
