DNA-Templated Synthesis-Assisted Libraries

DNA-templated synthesis is a general approach to create a variety of small molecules with DNA tags. The DNA-linked reagents could be brought into [Pg.264]

The strategy was used first by Naylor and Gilham to catalyze chemical phos-phodiester bond formation by hybridization of a poly(A) template and a poly(T) hexamer [19]. The method was developed by Liu and coworkers to tackle three issues (1) synthesis and selection of DNA-templated small-molecule libraries, [Pg.265]

In 2009, Hansen et al. have proposed another DTS-based library synthesis approach, in which three-way DNA hairpin-looped Junctions assist the library [Pg.265]

The ability of DNA-templated synthesis to support a variety of transition-state geometries suggests its potential in directing a range of powerful aqueous-compatible synthetic reactions. The apphcation of the strategy is not commonly used because of synthetic difficulties but could be a good method to build and discover small molecules with unique structures such as macrocycles. [Pg.266]
