Digestion preparation

Several protocols including in-gel digestion, preparing gel slices, and some others for MS analysis. 

Select slides with optimized proteinase K digestion and follow the steps below for DNase digestion. Prepare in an Eppendorf tube 1.0 pL RNasin (RNase inhibitor), 11.5 pL UV-irradiated dH20, 37.5 U DNase (RQl RNase free DNase, Promega Inc., Madison, Wl)/(1 U/pL) Total (per slide) 50.0 pL. 

These agents are administered to aid in the digestion of food. The primary digestant preparations contain pancreatic enzymes or bile salts. Pancreatic enzymes such as amylase, trypsin, and lipase are responsible for digestion of carbohydrates, proteins, and lipids, respectively. These enzymes are normally synthesized in the pancreas and secreted into the duodenum via the pancreatic duct. Bile salts are synthesized in the liver, stored in the gallbladder, and released into the duodenum via the common bile duct. Bile salts serve to emulsify lipids in the intestinal tract and are important in lipid digestion and absorption.

As an alternative to TCA precipitation, transfer 100 //I of the SI-digested preparation to a dry 2.4-cm Whatman DE-81 filter disk in a vial. Wash the disk twice with 5-ml volumes of 5% Na2HP04 and twice with 5-ml amounts of distilled water. Dry the filter and measure the radioactivity. 

The plot of CL intensity versus time presented in figure 6 shows the effect of varying the quantity of fresh autologous serum used as the source of complement. Zymosan, a boiled, proteolytically digested preparation of yeast cells, was used as the particle to be phagocytosed. Zymosan is well established as an activator of the alternative pathway of complement. Each vial contained an equivalent quantity of zymosan. The differences in the curves of CL therefore reflect the quantity of serum used for opsonification. 

A similar microwave digestion preparation procedure was used to determine the uranium content and its isotopic composition in soil samples collected in the vicinity of the Tokai-Mura plant in Japan after the criticality accident in 1999. The samples were oven-dried at 80°C to a constant weight and then powdered. Samples were placed in sealed Teflon pressure decomposition vessels with HNO3, HF, and HCIO4 and digested in a microwave oven, then evaporated to dryness and dissolved in 2% HNO3 prior to ICPMS analysis (Yoshida et al. 2001). This analytical method did not require separation on chromatographic columns. 

Iodine formed according to Eq. (10.37) is hnmediately reduced to iodide as long as there is any ascorbic acid present Thus, to successfully convert iodate to iodide, the sample must already contain iodide. Recovery data for iodate as iodide range from 80 to 127% for digests prepared at room temperature and from 94 to 103% for digests prepared at 70 °C.

Injection of an enzymatic digest of human insoluble skin collagen onto a HPLC showed a single major peak in the chromatogram when monitored by a LW fluorescence detector (Fig. 1). It was noted that the size of the peak from collagen digests prepared from age 10 and 47 year-old individuals with type 1 diabetes were comparatively more intense versus those from nondiabetic individuals of similar age Furthermore, peak area increased with chronological age and was considerably more intense in individuals with ESRD.
