Among the compounds that fall within this class are hydrocodone (e.g., Vicodin), oxycodone (e.g., OxyContin—an oral, controlled-release form of the drug), morphine, fentanyl, codeine, and related medications. Morphine and fentanyl are often used to alleviate severe pain, while codeine is used for milder pain. Other examples of opioids prescribed to relieve pain include propoxyphene (Darvon) hydromorphone (Dilaudid) and meperidine (Demerol), which is used less often because of its side effects. In addition to their effective pain-relieving properties, some of these medications can be used to relieve severe diarrhea (for example, Lomotil, also known as diphenoxylate) or severe coughs (codeine). [Pg.234]

A congener of isomethadone, propoxyphene (Darvon), was synthesized and introduced as a hoped-for nonaddicting, safer drug for medium severity pain. In view of present knowledge with this compound, the earlier claims that tolerance did not develop even on prolonged administration, and that doses in up to 50 times therapeutic range have not been fatal, stretches credulity to the utmost. The absence of any structural resemblance to morphine was even emphasized. [Pg.175]

Another example is propoxyphene [33]. The (2R,3S)-enantiomer, sold as the hydrochloride and once marketed as Darvon , is an analgesic. The 2S,3R enantiomer is an antitussive and was once marketed as Novrad . Today, neither is marketed in the United States. Darvon was removed in 2010 and Novrad removed about thirty years earlier. The scientists at Lilly who discovered these compounds were clever in their naming. NOVRAD is the mirror image of the name, DARVON. [Pg.175]
