The Schreiber research group, which had performed the exploratory studies on the RCM mentioned earlier in this section, also conducted a systematic investigation of the cyclization step in a DOS approach to macrolactones, exemplified by the representative compound 296 (Figure 11.25, site of ring closure, chemistry used and reagent for macrolactonization indicated). The optimized reaction conditions (TCBC, DIPEA, toluene, rt, 20 h) thus developed on solid phase were used to construct a small 36 compound pilot libraiy. Interestingly, the chirality of the linear precursors was not found to dramatically affect the macrolactonization, which stood in contrast to the results with RCM substrates where a significant stereochemical bias had been observed.
