Cohesive-end method

This cohesive-end method for joining DNA molecules can be made general by using a short, chemically synthesized DNA linker that can be cleaved by restriction enzymes. First, the linker is covalently joined to the ends of a DNA... [Pg.249]

DNA fragments can be joined (G3) to create artificially recombinant molecules. There are currently three methods for joining DNA fragments in vitro. The first of these capitalizes on the ability of DNA ligase to join covalently the annealed cohesive ends produced by certain restriction enzymes. The second depends upon the ability of DNA ligase from phage T4-infected E. coli to catalyze the formation... [Pg.215]

This chapter jnst devotes to snch problems. This chapter describes how one can reveal ion-radical conversions by means of physical methods and kinetic approaches and on purely chemical information, inclnding data on material balance and the nature of end products. These methods are woven together to form a cohesive nnit. All the methods are considered separately, and then complex approaches are described in their applications to several representative reactions. [Pg.205]

It would be desirable to have simple tests capable of characterising the fluidisation behaviour or flowability of particulate materials on the basis of their bulk properties. To this end, Carr19 developed a system to characterise bulk solids with respect to flowability. Table 6 summarises the properties which are determined. In Carr s method a numerical value is assigned to the results of each of these tests, and is summed to produce a relative flowability index for that particular bulk material. Given the extensive use of these empirical techniques in academia and industry, a brief review on the subject is reported here. Nevertheless, it should be emphasised that these techniques allow measurements of the flow-ability or cohesion of materials solely in their stationary or compressed status and at ambient conditions. A direct relationship between these... [Pg.227]

Compressed solids, tablets, or caplets, are prepared by placing the blend of component additives in a cylinder or die, above a moveable piston or punch. An upper punch is brought into the top of the piston, and pressure applied to the distal ends of the punches forces the powder into a compact (Fig. 7). The quality of the product depends upon the cohesive forces acting on the powder upon compression. These cohesive forces are influenced by the selection of additives in the dosage formulation. One method of evaluating... [Pg.3905]
