Clomiphene

For the purposes of this article, antiestrogens are compounds that counteract the biological activity of estrogens at the receptor level. In the late 1970s, there were no steroidal antiestrogens in widespread clinical use. Clomiphene [911 -45-5]( ) and tamoxifen/7(954(9-25 -/7(9) were nonsteroidal antiestrogens that had been employed for the treatment of female infertility and breast cancer, respectively. [Pg.232]

Alkylation of 4-hydroxybenzophenone by means of base with 2-ihlorotriethylamine affords the so-called basic ether (29). In a lequence analogous to the preparation of 28, the ketone is first reacted with benzylmagnesium chloride (30). Dehydration (31) and rlilorination complete the synthesis of clomiphene (32). ° It is itf note that the commercial product is in fact a mixture of the... [Pg.105]

As noted previously, triarylethylenes substituted by a basic group, such as clomiphene, exhibit estrogen antagonist activity. Formal cyclization of that molecule to a more steroid-like, rigid conformation enhances potency in this series. [Pg.148]

Clomiphene (Clomid) is a synthetic nonsteroidal compound that binds to estrogen receptors, decreasing the amount of available estrogen receptors and causing the anterior pituitary to increase secretion of FSH and LH. It is used to induce ovulation in anovulatory (nonovulating) women. [Pg.511]

Administration of clomiphene may result in vasomotor flushes (which are like the hot flashes of menopause), abdominal discomfort, ovarian enlargement, blurred vision, nausea, vomiting, and nervousness. HCG administration may result in headache, irritability, restlessness, fatigue edema, and precocious puberty (when given for cryptorchism). [Pg.511]

These drug are contraindicated in patients with known hypersensitivity to tire drug. Clomiphene is contraindicated in patients with liver disease, abnormal bleeding of undetermined origin, or ovarian cysts, and during... [Pg.511]

CLOMI PH EN E Clomiphene is an oral tablet prescribed for 5 days and is self-administered in the outpatient setting. [Pg.514]

Judy Cowan, age 28 years, has been prescribed clomiphene to induce ovulation and pregnancy. Judy is very anxious and wants desperately to become pregnant. Her husband, Jim, has come to the clinic with her. Discuss assessments the nurse would consider important before initiating treatment with clomiphene. Discuss information the nurse would include in a teaching plan for Jim and Judy. [Pg.529]

Which of die following adverse reactions would die nurse expect with die administration of clomiphene ... [Pg.529]

Some tumors are estrogen-dependent and the use of an antiestrogen has therapeutic value. One such antineoplastic agent appears to be patterned after clomiphene. Complex aryl ketone 204 is treated with phenyl magnesium chloride and the resulting tertiary... [Pg.127]
