Cisplatin resistant model

Dimeric complexes like [Cl(NH3)Pt H2N(CH2)4NH2 Pt(NH3)Cl]Cl2 are also being investigated as they bind to DNA in a different way to that involved in cisplatin binding and are active in cisplatin-resistant human tumour cells. In addition to the extensive oral antitumor studies described above in mice bearing the ADJ/PC6 plasmacytoma and human ovarian carcinoma xenografts, JM216 has also been evaluated in vivo in two murine models of acquired cisplatin resistance. Schedule-dependency effects have also been determined. 

At the preclinical level, oral antitumor activity has been observed with two other classes of platinum compound, the PtIV monocarboxylate, C(5)-OHP-C1, and the first Ptn complex to exhibit oral activity, the sterically hindered AMD473. Now licenced to Zeneca, AMD473 exhibited promising circumvention of acquired cisplatin resistance against both in vitro and in vivo preclinical models.
