These results may be explained by taking into account an hypothesis for the fate of oleuropein in vivo. If oleuropein is degraded to hydroxytyrosol and, according to the results reported by Bai et al. [98], this metabolite is rapidly removed from the plasma, then at the time interval of 2 hrs, when plasma was collected in the study by Speroni et al [96], the anti-oxidant molecule would not be present any longer, thus explaining the lack of effect of plasma of oleuropein treated rats on the chemiluminescence reaction. In contrast oleuropein or its metabolites might be eliminated through bile, which responded positively to the test. 

The analysis of a molecule with one site of action is a good indicator for the LD50, such as in phenacyl alcohol derivatives. Steroids, however, are complex molecules, and their metabolites contribute to the experimentally measured LD50 values. If the toxicity can be related to superoxide, the measurement of chemiluminescence intensity can be used for a drug candidate screening. Further studies of the substituent effect of steroids are required before LD50 values can be estimated from computational chemical calculations. 

The ultimate chemical carcinogenic metabolite of the parent carcinogen benzo[a]pyrene has been shown to be the 7,8-dio1-9,10-epoxide (DE) of benzo[a]pyrene produced from the 7,8-diol precursor. The mechanism study has shown that the observed microsomal chemiluminescence of benzo[a]pyrene arises mainly from the dioxetane of the 7,8-diol.
