Cell mutants

The recent synthesis of (—)-tetracycline by Myers and co-workers incorporates a biocatalytic step in the first stage which oxidizes benzoic acid aerobically to an a,f)-dihydroxy derivative in the presence of a whole-cell mutant strain of Alcaligenes eutrophus. Figure 4.61 shows a reduced tree diagram for the synthesis and Table 4.28 summarizes the metrics parameters.

The assignment of obtaining of cell mutant cultures of domestic animals appears to be one of the hybridization problems. The cultures grown at different time are preserved in Animal Cells Collection and in the cryobank of the Institute of experimental veterinary, and the selection of TK- or GPRT-defective cells needed the answering of several questions.

Still another approach to solving the problem of detecting intenne-diates in the assembly of the lipid-linked oligosaccharide comes from the use of cell mutants having defects in the synthesis of an essential intermediate. This approach has proved productive (see later).

A fourth type of WgaR CHO cell mutant has now been isolated (Stanley, manuscript in preparation). This mutant is more highly resistant to WGA than the previously described mutants and it has been shown to belong to a new complementation group (group VIII). 

In this paper, the glycolipids of WgaRvm cells are compared with those of parental CHO cells and the other WgaR CHO cell mutants.
