Deoxyfluoro- and deoxy-sugars are useful in studies both of binding interactions between carbohydrates and proteins, and in mechanisms of glycosyl transfer. This paper describes how we have used such analogues to measure hydrogen bond strengths in a carbohydrate/ protein complex of known structure. It proceeds to review how such substitutions affect the rates of glycosyl transfer in a non-enzymatlc model reaction and then applies this Information to the results obtained in our enzymatic system.

Mass spectrometry (FAB, MALDI, and ESI modes of ionization) are used extensively to monitor and confirm products of chemical synthesis. Unfortunately, important details such as the type of MALDI or FAB matrix or the type of instrument used to record the spectra are often missing from these publications as the techniques tend to be regarded as routine. There are a few areas—for example, analysis of glycodendrimers and carbohydrate-protein complexes— where the high-mass capability of MALDI is particularly useful.
