2-Bromo-2-nitro-propan

Another commonly used bactericide is 2-bromo-2-nitro-propane-1,3-diol.

Nitropropane (178 g., 2.0 moles) is added with cooling to a solution of 132 g. (2.0 moles) of 85% potassium hydroxide in 500 ml. of water. The solution is cooled in an ice bath and stirred while 320 g. (2.0 moles) of bromine is added. (Hood.) The rate of addition of the bromine is such that the temperature of the reaction mixture is held in the range 35-40°. Sufiicient solid sodium sulfite is added to the red solution to obtain a colorless mixture, and the heavy oily layer is separated and washed with water. After drying over sodium sulfate, the product is distilled to give an 89% yield of 2-bromo-2-nitro-propane, boiling at 151.7-151.8°/745 mm.

Bronidox is chemically the formal of 2-bromo-2-nitro-propane-l,3-diol (Bronopol) and therefore more stable than Bronopol. Figure 23 demonstrates that formaldehyde is released from Bronidox in a small amount only and very slowly even at pH 9. However, finally the degradation pathway follows the Bronopol pathway leading to formaldehyde, bromide, nitro-alcohols and nitrite.

In cosmetics, use of 5-bromo-5-nitro-l,3 dioxan (Bronidox) [II, 17.15.] and 2-bromo-2-nitro-propane-l,3-diol (Bronopol) [II, 17.14.] has declined dramatically, as there exists the risk that in cosmetic formulations containing amines N-nitroso compounds may originate which are known as carcinogens.
