To prepare HaloTag blocking ligand, HaloTag Succinimidyl Ester (04) Ligand (20 mM) was incubated with 100 mM Tris-HCl (pH 8.0) for 60 min at 25°C to mask the functional groups. 

The cells were continuously incubated in the growth medium including a different amount of HaloTag blocking ligands (500 nM, 5 pM, and 50 pM) for the indicated time period (Chase) except for the sample at the zero time point (Fig. la). 

The TMR-labeled HaloTag proteins were observed and recorded in the cultured cells using the fluorescent microscope BioZero in the growth medium with or without 5 pM blocking ligand at the time points indicated in Fig. lb.
