Biosynthesis production kinetics

The selection of appropriate production strains for PHA biosynthesis from lignoceUulose-derived substrates mainly depends on the conversion rales of hexoses and pentoses by the organism. If the different sugars are not used in parallel and with similar rales, the bioprocess to be developed will be rather compUcaled. Sugars that are not accepted as a substrate by the strain or that are utilized considerably more slowly than others can pile up in the fermentation broth and may then cause inhibitory effects that are very likely to negatively influence growth and production kinetics and yields. [Pg.107]

Because this enzyme catalyzes the committed step in fatty acid biosynthesis, it is carefully regulated. Palmitoyl-CoA, the final product of fatty acid biosynthesis, shifts the equilibrium toward the inactive protomers, whereas citrate, an important allosteric activator of this enzyme, shifts the equilibrium toward the active polymeric form of the enzyme. Acetyl-CoA carboxylase shows the kinetic behavior of a Monod-Wyman-Changeux V-system allosteric enzyme (Chapter 15). [Pg.806]

Irradiation of 2,2-dimethyl chromene through Pyrex using a 550-W Hanovia lamp initiates a retro 4 + 2 reaction to form the extended quinone methide 4, which reacts with methanol to form a pair of methyl ethers (Scheme 6A).18 Flash photolysis of coniferyl alcohol 5 generates the quinone methide 6 (Scheme 6B) by elimination of hydroxide ion from the excited-state reaction intermediate.19 The kinetics for the thermal reactions of 6 in water were characterized,20 but not the reaction products. These were assumed to be the starting alcohol 5 from 1,8-addition of water to 6 and the benzylic alcohol from 1,6-addition of water (Scheme 6). A second quinone methide has been proposed to form as a central intermediate in the biosynthesis of several neolignans,21a and chemical synthesis of neolignans has been achieved... [Pg.44]

Although cell-free synthesis of 6-dEB was achieved, kinetic studies demonstrated that the process was very inefficient [34], The low rate of biosynthesis was likely due to the fact that the association of the three DEBS proteins in vitro was suboptimal. To simplify in vitro synthesis and to facilitate mechanistic analysis, a truncated version of the erythromycin PKS was created. The protein, DEBS 1-TE, was engineered by relocating the thioesterase (TE) from the end of DEBS 3 to the terminus of DEBS 1 (Fig. 9a,b) [35], In vivo, this bimodular construct synthesizes two triketide lactone products, one derived from propionate as a starter unit, and the other from acetate. [Pg.439]

In rats equilibrated with radioiodine-labelled T4 or T3 roughly half of the radioactivity appears as I- in the urine and the other half as free iodothyronines in the feces [12]. Treatment of the rats with 6-propyl-2-thiouracil (PTU) results in a marked decrease in urinary radioactivity and a reciprocal increase in fecal clearance [12]. Also, in humans, PTU has been shown to inhibit peripheral iodothyronine deiodination besides its well-known effect on thyroid hormone biosynthesis [13]. Compared with the rat, deiodination is an even more important pathway for the clearance of thyroid hormone in man as evidenced by the greater proportion undergoing urinary clearance [2]. Furthermore, estimation of iodothyronine turnover kinetics in humans has demonstrated that a major fraction of T4 disposal is accounted for by plasma production rates of T3 and rT3 [2,3],... [Pg.83]

J. J. Hopfield has proposed a so-called energy relay model [101], in which the ping-pong mechanism operates on two identical substrate molecules and releases two identical product molecules. The model introduces the novel idea of dynamic cooperativity by which biomolecular processes in living cells, such as DNA replication and protein biosynthesis, can achieve high fidelity. The model was developed as an alternative to the kinetic proofreading mechanism which we shall discuss in Chapter 5. [Pg.96]

The loss of the relatively C-enriched carbon atoms C-3 and C-4 of glucose [107], and the kinetic isotope effect on the pymvate-dehydrogenase reaction /7 0(5/ in the biosynthesis of acetyl-CoA have for consequence that isoprenoids from the MA-pathway are depleted in relative to carbohydrates from the same source by 4 to 6%o, whereas this C-depletion attains in products from the DOX-pathway only 2 to 4%o. C-pattems of the two types of isoprenoids, which would certainly be different, are still not yet available. [Pg.631]
