A number of methods for the selection of bacteriophage displaying V gene products by affinity purification have been described including ... [Pg.456]

RNAs produced in vitro are useful in the development of in vitro splicing systems where the RNA produced is a primary transcript. In addition, the RNA can be used as a template for in vitro translation, and in mapping exons tuid introns in genomic DNA. Genes under control of bacteriophage promoters can also be expressed in bacterial cells to facilitate expression and/or purification of the proteins they encode. [Pg.125]

SmaU-sized plasmid DNA (less than 20 kb) easily can be purified with standard techniques, and an efficient protocol is shown in Protocol 9. High-quality PI bacteriophage, BAC, and even YAC DNA for micro-injection can be purified from preparative agarose pulsed-field gels. This procedure is somewhat more complicated than handling small-sized plasmid DNA but can easily be performed with some experience. A protocol for purification of PI bacteriophage and BAC DNA is shown in Protocol 10. [Pg.157]
