Bacillus megaterium protoplasts

Spiegelman, Aronson, and Fitz-James found that digestion of protoplasts of Bacillus megaterium led to the liberation of nuclear bodies of the protoplasts. Such bodies were collected by centrifugation for 5 min. at 10,000 x g. 

Beckett and Robinson i have conducted studies with whole cells, protoplasts, and lysed protoplasts of Bacillus megaterium and shown that the site of reduction of nitrofurazone is the cytoplasmic membrane rather than the cell wall or the cytoplasma.

Bacillus subtilis transformants were identified by their resistance to naphthalene, parachlorobiphenyl, or dibenzofuran. Controls subtilis protoplasts regenerated without the addition of B. megaterium plasmid DNA) exhibited no increased reistance to these three compounds. That each transformant was B. subtilis and not a contaminant was verified by testing for the B. subtilis chromosomal markers. Only streptomycin resistant strains that showed arginine and leucine dependent growth were used in subsequent experiments.
