To improve production of rhamnogalacturonase by Aspergillus aculeatus CBS 115.80 shake flask ejqjeriments were performed on several substrates. Cross reactivity was found after transfer to thamnose in combination with galacturonic acid and on apple pectin, citrus pectin, beet pectin and sugar beet pulp. No cross reactivity was found after transfer to meda containing simple carbon sources such as sucrose, glucose, fiuctose, rhamnose or galacturonic acid. 

Addition of fermentation broth fi-om Aspergillus aculeatus CBS 115.80 to the isolated substrate resulted in an extinction increase measured by DNS-method.. 

RG-lyase was purified from Pectinex Ultra SP-L, produced by Aspergillus aculeatus, using anion- and cation-exchange chromatography. The purified RG-lyase differed from RG-hydrolase in pi and pH optimum and stability (Table I).

CE 12 contains serine enzymes which are likewise deacetylases. The rham-nogalacturonan I acetyl esterase of Aspergillus aculeatus removes acetyl groups from the 2- and 3-positions of the main-chain galacturonic acid residues of rhamnogalacturonan I it is a serine esterase, with a Ser-His-Asp catalytic triad and an oxyanion hole made from two main-chain peptide NHs and an Asn side-chain, but does not have the ot/p fold.
