Antifungal drugs case study

Few randomized controlled studies have been performed for antifungal drugs because of the difficulty in recruiting a sufficient number of cases within a given time frame. Evidence-based information is scant, as many of the reports have been in the form of single case reports, studies of small numbers of patients, or retrospective reviews of patient records (Table 11-13). 

Antifungal drugs Nystatin and natamydn are toxic to the retina and should not be given intravitreally. Voriconazole seems to be non-toxic according to in vitro studies and several case reports however, blurred vision, halos, altered color perception, and visual hallucinations can occur. 

Taupitz et al. [24] have reported a case study of the potent antifungal drug, itraconazole, which has a poorly soluble weak base which precipitates in the small intestine, its binary and ternary complexes and marketed formulation (Sporanox ). All these formulations were subjected to solubility and dissolution studies in media mentioned in Table 12.5 at 37°C. All the formulation approaches enhanced the solubility of drug in all media but more pronounced results were observed for ternary complexes (itracon-azole-CD-soluplus ), which are tabulated in Table 12.5.
