Although several types of fluorescent beads were proposed as a microscopic fluorescence standard 30 years ago,2 beads have not been used as a proteinembedding matrix for routine IHC on FFPE tissue. We recently tested primary coated beads ( Dynabeads, Dynal, New York) that are coated with a goat anti-mouse antibody on the surface of the beads. Primary antibody mixture of rabbit pan-Cytokeratin antibody (Santa Cruz) (1 50 dilution) for carcinoma cells and rat anti-F4/80 (16) (1 25 dilution) for macrophages. Store the undiluted pan-Cytokeratin antibody at4°C never freeze. The undiluted F4/80 antibody should be stored at -20°C in several aliquots prevent frequent freeze-thaws. Cytokeratin cocktail antibodies can be used to identify MECs (in addition to CK14 and CK17). However, because of the proximity of MECs to acinar cells, differentiation is difficult because these cocktails also immu-nostain acinar cells. Anti-smooth muscle actins react with stromal myofibroblasts in addition to MECs " and thus are not specific for MECs. The cross-reaction with myofibroblasts makes it difficult to identify MECs specifically, especially for ductal carcinoma in situ (DCIS) cases in which periductal stromal desmoplasia may exist. [Pg.764]
