It is now possible to image not only postsynaptic, but pre-synaptic and intrasynaptic neurotransmission (Fig. 58-5). Presynaptic sites, such as the dopamine transporter and the serotonin transporter the presynaptic dopamine vesicular transporter (VMAT-2) and the acetylcholine transporter extrasynaptic sites such as the enzymes which break down neurotransmitters, e.g. MAO A and MAO B with radioligands that bind to post or pre-synaptic sites, i.e. dopamine competing with radioligands such as UC raclopride (see Fig. 58-9) (PET (Fig. 58-10) can be measured under basal conditions or following drugs which either decrease (e.g. AMPT) or increase (e.g. intravenous amphetamine) intrasynaptic dopamine.

In 1984, the AMPTE mission launched the first carbon-foil TOF-MS into space, which would have been the second, had the Challenger shuttle disaster not delayed the Ulysses launch until 1991 (Fig. 11.2) [23]. The photons were filtered out by a traditional blackened deflection system, which directed the ions toward the 2 p,g/cm2 thick foil mounted on an 85% transparent grid almost a square centimeter in area. The grid provided the support needed to survive the launch. The foil thickness permitted >2keV/nuc ions to pass through and hit a SSD some 10 cm away. To ensure that the ions made it through the foil and also through the dead layer on the SSD (caused by the upper electrode), the foil and the entire TOF section were floated at 20 kV to post-accelerate the ions. Electrons sputtered off the carbon foil became the start, whereas electrons sputtered off the SSD became the stop pulse for the TOF.

Other drug effects that decrease DA activity also support this position. Thus, when DA synthesis is blocked by a-methyl- p-tyrosine (AMPT), the dose necessary for an antipsychotic effect is reduced (i.e., the dose-response curve is shifted to the left by the interaction between dopamine and AMPT). A drug such as reserpine that can deplete DA stores also has relatively mild antipsychotic properties. Also, aripiprazole, which has D 2 presynaptic agonist properties, decreases the production of DA, as well as blocking D2 postsynaptic receptors.

Downward Movement—In the case of downward movement, it is necessary to adjust the driving force in Eq (15-5) to take care of the capillary forces. Assume, following Green and Ampt, that this added driving force is taken as an extra head, hCi so that the total driving force is gp(h + hc) instead of ghp. Also, since the water occupies the pore-space of the soil, it follows that at any stage the velocity of the water front will be given by the equation... 

Green and Ampt (1911) tested this equation for a soil and found the L2/t to be constant only after several hours. They found better agreement for the same data by using the expression...
